Currently, ASCEM has established the structure determination pipeline by single particle analysis. Due to lack of expertise in tomography, ASCEM does not provide ECT (electron cryo-tomography) service at this moment. However, we are willing to connect you with our overseas ECT advisory experts for potential collaboration while ASCEM is working on the development of ECT.
An Overview of Application Procedures
 
1. To increase effectiveness of obtaining high resolution structure data, ASCEM’s equipment is principally planned to be applied step by step.
2. CEM1 is an electron microscope operating under room temperature. CEM2/3/4 are different ranks of CRYO-electron microscopes, and Vitrobot is the equipment for sample preparation.
  Usage order CEM1 (negative staining) → (Vitrobot) → CEM2 → CEM3 → CEM4.
3. Training and certification are necessary before applying for Vitrobot.
4. CEM3 will be opened for night time or holidays in the future, operating by super-user only.
 
Equipment Application and Reservation
Vitrobot、CEM1、CEM2
  General samples: contact Facility Managers for discussion and then complete the online reservation via Academia Sinica Facility Reservation System.

  Biohazardous samples: submit a proposal via Google Form, along with related certificates of biosafety.
CEM3、CEM4
  1. Submit a proposal via Google Form.
    A protein complex will be considered as one project (i.e. one Google Application Form). Different ligand/complex will be a different project (separate Google Application Form and project number), but the relevance among projects can be described in the Google Forms.
    After submitting a Google Form, a Project Number with 10 digital numbers will be automatically produced by referring to submission time.
    Whenever finishing data collection one time, a Manager Report will be provided to Applicant. After Applicant submits a User Report, a continuous application for another data collection can be proposed by the same Project Number (10 digits).
  2. Every application will be reviewed by ASCEM Priority Committee Team (PC Team). Once approved, contact Facility Managers for time assignment.

  3. Based on different duties of every ASCEM staffs, Facility Mangers will not view the Google Forms, so please assist Managers to understand your project for an appropriate service.
 
Charge & Closure
  1. Every case will be charged in accordance to the record on the Academia Sinica Facility Reservation System. The PI/Supervisor will receive a confirmation request email for every case closure.
  2. The Applicant shall remind your Supervisor/PI to complete the case closure confirmation for individual cases. Then, ASCEM can monthly compile all the closed cases and send a payment notice to Supervisor/PI.
    Academia Sinica (AS) budget users: charged via intramural budget accounts.
    Non-AS academia users: paid via non-AS budget payment procedures.
  3. If the payment source comes from non-AS projects, the rate of charges will follow the non-AS academia users according to the official document, No.1090502588, announced by Academia Sinica on 2020/04/10.
 
Workflow Chart
 
NOTE 1:
For applying CEM3 and CEM4, or when dealing with virus or other biohazardous samples, please submit a proposal via Google form (Workflow Chart STEP 1). If the proposal is approved, the Facility operation time will be assigned by Facility Managers. For virus or other biohazardous sample, please read the Requirements and Affidavit as well as be ready for the necessary data before application.

NOTE 2:
The first time facility applicant is highly recommended to apply an account and get facility authorized (Workflow Chart STEP 3) soon after the approval of project or confirming with Facility Managers for speeding up the reservation procedures afterwards. Only after completing the online Reservation System (Workflow Chart STEP 4), the user is officially allowed to operate the applied facility.

NOTE 3:
a) For applying CEM1, CEM2, and Vitrobot as well as general samples, a request can be directly submitted via Academia Sinica Facility Reservation System (Workflow Chart STEP 4) after discussing and confirming the Facility time assignment with Facility Managers except that CEM2 / Vitrobot certified users may skip the prior discussion. In addition, CEM1, CEM2, and Vitrobot are limited to users within Academia Sinica.

b) “ATTENTION” From 2019/01/01, the Academia Sinica Facility Reservation System – “Cryo-EM Core Facility, IOC” and “Cryo-TEM Core Facility, ICOB” will be closed for access. Application of CEM1 (JEOL JEM-1400), CEM2 (FEI Tecnai G2 F20 TWIN), and Vitrobot will be shifted to “Academia Sinica Cryo-EM Center (ASCEM)”. In addition, for those who plan to use ASCEM facilities in 2019 may now submit requests via “Academia Sinica Cryo-EM Center (ASCEM)” after reading detailed procedures on ASCEM “Access” webpage.

* According to the facility cost analysis formula of Division of Instrument Service of Academia Sinica (AS), the cost per day is respectively NTD100,000 and NTD140,000 for CEM3 and CEM4. ASCEM has been giving CEM3/4 academia users 90% discount of service charges since operation started under the significant financial subsidization from AS and Taiwan Protein Project (TPP). However, in response to the new policy of AS and the budget reduction of TPP since 2020, ASCEM has to adjust service rate of CEM3/4. While AS budgets users remain the benefits of 90% discount of service charges as before, the charges for non-AS academia users is adjusted to 65% off.

ASGC Annual Data Storage Fee: NTD 1,000/TB per year. From September to December of 2019, only 1/3 of the annual fee will be charged, and the unit is NTD333/TB. (Start from 2019.09.01)
