Currently, ASCEM has established the structure determination pipeline for single particle analysis. ECT (electron cryo-tomography) service is still under development, so no routine service is provided at this moment. For any questions, please contact us via the EMAIL cryoem@gate.sinica.edu.tw.
An Overview of Application Procedures
 
1. To increase effectiveness of obtaining high resolution structure data, it is recommended to follow the following sequence:
  CEM1 is an electron microscope operating under room temperature. CEM2/3/4 are different ranks of CRYO-electron microscopes, and Vitrobot is the equipment for sample preparation.
  Recommended usage order: CEM1 (negative staining) → (Vitrobot) → CEM2 → CEM3 → CEM4. Please select the most suitable instrument according to the sample characteristics. For consultation, please contact us via cryoem@gate.sinica.edu.tw.
2. Training and certification are required before applying for Vitrobot. When necessary, users may apply for the staff operation (limit to use Vitrobot2). The fee will be additionally charged and 4 cryo-grids production per time in maximum. Please fully discuss with the staff before submitting a “Service Request” online. For more details, please refer to CryoSample/Data and Regulations of Vitrobot.
3. For viruses, even inactivated viruses, or other biohazardous samples, please submit your application via the Google form (including Vitrobot) and the Requirements and Affidavit should be attached.
4. Certified independent users can use the facility during working days from Monday to Friday 09:00-20:00, excluding national and public holidays.

  5. Resolutions for CEM4 Overload
  6. CEM3 is reserved one day every week (Tuesday preferably) for TPP promotion cases.
  7. The above policy is currently not applicable to tomography experiments.
 
Charge & Closure
  1. Every case will be charged according to the record on the Academia Sinica Facility Reservation System. The PI/Supervisor will receive a confirmation request email for every application.
  2. The applicant shall complete the “Time and Charge Confirmation” for each individual case. Then, ASCEM will compile all the closed cases monthly and send a payment notice to Supervisor/PI.
    Academia Sinica (AS) charge rate: charged via intramural budget accounts.
    Non-AS academia charge rate: paid via non-AS budget payment procedures.
  3. If the payment source comes from non-AS projects, the rate of charges will follow the non-AS academia users rates, according to the official document, No.1090502588, announced by Academia Sinica on 2020/04/10.

Agreement for the Regulations of Vitrobot
  All Vitrobot Users need to sign for this Agreement, and then please bring the original document to Vitrobot Manager/Staff (see Agreement No. 7) on site at the first time of operation.

* According to the facility cost analysis formula of Division of Instrument Service of Academia Sinica (AS), the cost per day is respectively NTD100,000 and NTD140,000 for CEM3 and CEM4. To promote the research and the application of cryoEM, ASCEM has been giving CEM3/4 academia users 90% discount of service charges since operation started under the significant financial subsidization from AS and Taiwan Protein Project (TPP). However, in response to the new policy of AS and the budget reduction of TPP since 2020, ASCEM has to adjust service rate of CEM3/4. While the payment using AS budgets remains the 90% discount of service charges as before, using non-AS budgets is adjusted to 65% off. Meanwhile, considering that the off-AS researchers usually are funded by non-AS budgets only and may lack of cryoEM professional manpower in time or have limited budgets and so on, TPP has developed a cryoEM platform with a professional research team for supporting those in need to apply and take advantage of the resource.

***The charge scheme is scheduled to be raised on April 1, 2021 in accordance to the consensus obtained in the ASCEM Reorganization Plan & User Meeting held on December 29, 2020.
